Overview
Retrovirus has been widely used to transduce foreign DNA into dividing mammalian cells. FenicsBio’s Retrovirus Production Service is a one-stop service covering gene synthesis, generation of retrovirus vector, retrovirus packaging, concentration, purification, and titration.
We have developed innovative technologies for concentration and purification of the retrovirus supernatant. Our purified retroviruses are serum free, pure enough for in vivo experiments.
Except the small-scale production, all retroviruses are concentrated, purified and functional titrated on mammalian cells. Depending on the markers on the retrovirus constructs, the titration readout can be counting antibiotic resistant cell clones, counting fluorescent positive cell clones or genomic real-time PCR.

Project Initiation
If you have retrovirus DNA construct, please send us at least 1ug of the plasmid DNA, and plasmid map/sequence. Our procedures will include:
- Retransformation and preparation of endotoxin-free plasmid
- Transfection of HEK293T cells with your plasmid and retrovirus packaging plasmid
- Collecction of retrovirus supernatant
- Virus concentration and purification
- Functional titration on mammalian cells
- Aliquot and shipping
If you need us to generate retrovirus constructs, please send us detailed requirements for the construct, such as promoter to use, antibiotic selection gene, reporter et al. We can generate retrovirus constructs from scratch or based on the material you have.
For cloning cDNA, please send us at least 10 ug of the plasmid containing your target cDNA or the sequence of the cDNA for gene synthesis.
For shRNA/sgRNA, please send us the sequence if you know the sequence that works well, or we can design shRNA/sgRNA for you.
