Overview

Knockout cell is a powerful tool for studying gene functions, molecular mechanism of disease pathogenesis and discovering drug target. But generation of knockout cells needs optimization of lots of experimental conditions and is time-consuming. FenicsBIO provides one-stop Cell Line Knockout service from sgRNA design to knockout cell line validation and assay development. We take of the whole cell line knockout process so that you can use your valuable time for more important discoveries.

Our cell line knockout service uses a set of three sgRNAs dispersed with optimal distances at the starting of cDNA, which cause a large deletion easy to be identified by In-del PCR screening, ensuring complete knockout of protein expression.  We provide services for generating pooled knockout cell line and clonal knockout cell line. For pooled knockout cell line, at least 50% of the pooled cells are knockout cells, which can be used directly for functional assay or proceed to clonal selection. For clonal knockout cell line, two individual knockout cell clones will be provided.

Deliverables/Lead Time

Deliverables:

  • Pooled knockout cell service:

One vial of knockout cell pool at 1x10^6 cells/vial and one vial of control parental cells at 1x10^6 cells/vial will be delivered.

  • Clonal knockout cell service:

One vial for each of the two knockout cell clones at 1x10^6 cells/vial and one vial of control parental cells at 1x10^6 cells/vial will be delivered.

Lead Time:

  • Pooled knockout cell service: 4-5 weeks depending on the cell types.
  • Clonal knockout cell service: 8-10 weeks depending on the cell types.

QC Testing

  • Sanger sequencing
  • Mycoplasma test
  • (Optional) RT-PCR, Western blot, FACS

Project Initiation

Phase I: Project requirement information collection

Our scientists will work with you to figure out the details of your cell line knockout requirement, including the gene ID you want to knockout, parental cell line, whether you want all allele knockout or single allele knockout, whether you want pooled knockout cells or clonal knockout cells, whether you need optional knockout cell line verification, such as RT-PCR, Western blot, FACS.

Phase II: Project plan approval

Based on the above information you provided, we will formulate a project plan and send you a quotation. After you approve the project plan, you can use the quotation to place an order.

Phase III: Starting material shipment

We prefer customers to send us the parental cells if possible although we can buy it on your behalf sometimes.

If you need us to do Western blot or FACS verification, please send us a workable Western blot or FACS antibody or we can buy it for you.
